The story of the endocannabinoid system begins in 1992 at Hebrew University in Jerusalem. Dr. Raphael (Ralph) Mechoulam — already famous for isolating and synthesizing THC in the 1960s — and his team made a groundbreaking discovery. They identified anandamide, the first known endocannabinoid, and uncovered a vast receptor network within the human body.

This was a turning point in modern biology. Scientists realized that our bodies naturally produce cannabis-like molecules and that these molecules interact with specific receptors to regulate critical functions. The ECS wasn’t just a curiosity — it was a central regulatory system that had gone unnoticed for decades.

The Core Components: CB1 and CB2 Receptors

At the heart of the ECS are two primary types of receptors: CB1 and CB2.

CB1 receptors are found mostly in the brain and central nervous system. They play a major role in regulating mood, appetite, memory, pain sensation, and motor control. This explains why THC, which strongly binds to CB1 receptors, can alter mood and perception.

CB2 receptors are located primarily in the immune system and peripheral tissues. They’re involved in controlling inflammation, immune response, and tissue repair. Phytocannabinoids: Plant-Based Partners

Cannabis plants produce more than 100 different cannabinoids, known as phytocannabinoids. Some of the most well-known include THC, CBD, and THCa.

THCa is the acidic precursor to THC and is found in raw cannabis flower. When heated — through smoking, vaping, cooking, or baking  — THCa is converted into THC, an “activated cannabinoid” that binds directly to CB1 receptors and produces psychoactive effects.

CBD, on the other hand, doesn’t bind strongly to CB1 or CB2 receptors. Instead, it influences the ECS indirectly, enhancing the body’s ability to regulate itself. This is one reason CBD is widely studied for its potential calming, anti-inflammatory, and neuroprotective effects without the high.
